Redefining

Three
breakthrough instruments were introduced that take mass spectrometry (MS)

On-the-Fly
Retention Time Shift Correction for Multiple Targeted Peptide
Quantification by LC-MS/MS
Selected reaction monitoring (SRM)
using triple quadrupole mass spectrometers is a powerful technique for
targeted protein quantification due to its high sensitivity and
selectivity. To satisfy growing demand for verifying large numbers of
putative targets found during the discovery phase, the instrument needs
to be capable of quantifying hundreds of peptides, with confirmation of
their identities, in a single LC-MS run. To address this challenge, a new
intelligent SRM (iSRM) technology has been incorporated into the Thermo
Scientific TSQ family of triple quadrupole mass spectrometers. This
technology enables processing of over 15,000 SRM transitions for
simultaneous quantification and identity confirmation of up to 1,000
peptides in a single LC-MS run.

LC-MS Analysis of Compounds for Early Drug Discovery: Quantitative and
Qualitative Screening in One Experiment
Full scan, ultra-high resolution and
accurate mass liquid chromatography-mass spectrometry (LC-MS) instruments
allow for rapid analyses of drug discovery samples without the need for
specific MS methods for target compounds. With the use of sophisticated
algorithms, Thermo
Scientific MetQuest software automatically processes the data
obtained from these instruments to identify compounds of interest at
mass-to-charge values that have not necessarily been pre-defined, while
allowing the re-interrogation of the same data set for the identity and
relative quantities of metabolites in both in vitro and in vivo
time course studies.

Acquisition for Rapid Quantitative and Qualitative Analysis Using the
Exactive Benchtop LC-MS High Resolution Mass Spectrometer
Current approaches to discovery stage
drug metabolism studies have focused on the use of targeted
analysis-based approaches, which necessitate the optimization of many
parameters while detecting only the specified compound. This application
note describes the use of high resolution, full scan MS, and Higher
Energy Collision Cell Dissociation (HCD) data obtained by the Thermo
Scientific Exactive LC/MS system for the quantitation and qualitative
analysis of a tolbutamide in vitro rat hepatocyte incubation. The
ability to do full scan acquisition for quantitation eliminates the need
for compound optimization, while enabling the detection of metabolites
and other non-drug related endogenous components.
